Pollen Formation and Germination of Colchicine - treated Intergeneric Hybrids between Brassica and Raphanus

Abstract
Large variations in pollen morphology and germination were observed in intergeneric hybrids between Brassica and Raphanus. Pollen fertility measured by stainability varied according to hybrids from different cross combinations. Hybrids between Brassica juncea¡¿Raphanus sativus ¢¥Jungkukcheongpi¢¥ and B. juncea¡¿R. sativus ¢¥Namwonmu had normal pollens, while those of B. campestris ¢¥Haruna¡¿R. sativus ¢¥Daibyosobudori¢¥ had no stainable pollens, indicating that large variations depend on the cross combinations. The rate of in vitro pollen germination in chromosome-doubled hybrids was 3.8 to 50.7% However, pollen tube growth was relatively poor in all germinated pollen. In vivo pollen germination and pollen tube growth on the stigma were good in almost all cross combinations studied, but the tip of pollen tube in style looked swollen, and the growth was stopped by the formation of callose plug.
